Spontaneous Diabetes Mellitus in the BB Rat



Abstract
The BB rat develops a diabetic syndrome that in many respects resembles human insulin-dependent diabetes mellitus. Although extensive evidence supports the role of autoimmunity in the pathogenesis of diabetes in the BB rat, the precise
immunologic
mechanisms leading to the beta cell destruction is not clearly defined yet. We studied the anti-islet cellular cytotoxicity in diabetic BB rats by comparing the effects of splenic lymphoid cells from diabetic BB rats and control rats on the
release
of
51Cr from damaged islet cells in vitro.
Splenic lymphoid cells for diabetic BB rats were meore cytotoxic to MHC-compatible Wistar-Furth(W/F) rat islet cells and a rat islet cell line than were lymphoid cells from control rats. This difference was amplified by incubating the lymphoid
cells for
24hr with 5§¶ concanavalin A (Con A). Splenic lymphoid cells were also cytotoxic to the YAC-1 cells, which are known traget for the natural killer (NK) cells.
These results suggest that anti-islet cellular cytotoxicity and NK cell activity are increased in BB rats.
